I would like to replace my Synology NAS with a notebook with VM and Xpenology. Can this enclosure be used for this with the current disks? With my backup NAS I have solved it with an enclosure from Orico (5bay), but I need more bays on the main NAS.

I don't know that directly. I use 2 of them with up to 8tb discs for storage spaces. Runs perfectly
