That shouldn't be a problem, space-wise. The first PCIe x16 slot is intended for dual-slot GPUs anyway, and after that there is another PCIe x1 slot in between. I would be more concerned about heat generation, since the two graphics cards are right next to each other. So good case cooling is a must.
